Iron Metabolism

The oxidation of iron in the substantia nigra region of the brain is among the core processes of neurodegeneration in Parkinson’s disease. Attenuation of oxidative conditions in the brain is a prerequisite to controlling iron metabolism and the preservation of the neuroprotective pigment neuromelanin.

Oxidative

Oxidative phosphorylation within mitochondria is fundamental to life. In this process, reactive oxygen species (ROS) are formed. While ROS’s play important roles as signaling molecules, their high reactivity and electrophilic nature must be kept within tightly regulated parameters. Parkinson’s disease features excessive oxidative stress and subsequent neuroinflammation.

Autophagy is an intracellular clean-up process, whereby misfolded proteins and cellular debris are degraded, removed or recycled. The glymphatic system modulates the removal of waste from the brain, including protein aggregates. Ubiquination is a related process, influencing what stays and what is degraded.

Cell Survival

The balance between cell survival and cell death is delicate. Both are dynamic and necessary for the health of the organism. The loss of nigrostriatal dopaminergic neurons is a core feature in Parkinson’s.
